Anti–paternal antibody detection in maternal serum. Method : Following isolation and extensive washing of paternal peripheral blood mononuclear cells, incubation with maternal blood serum is performed (crossmatch). In parallel, incubation with the paternal serum is performed as negative control. The cells are then washed and incubated with the following combinations of monoclonal antibodies (triple direct immunofluorescence – FITC/PE/ECD): CD3 – CD19 – IgM and CD3 – CD19 – IgG followed by flow cytometry analysis.
